Baird Parker Agar Base ISO is a selective and differential culture medium formulated for the detection and enumeration of coagulase-positive staphylococci in food, cosmetic and environmental samples. The base medium uses a mixture of meat/ casein hydrolysates, sodium pyruvate, glycine, lithium chloride and other selective agents to inhibit background flora while allowing staphylococci to grow.
Baird Parker Agar Base ISO (catalogue No. 1100) is a dehydrated agar medium intended for selective isolation and enumeration of coagulase-positive staphylococci from food, cosmetics, environmental or other materials. The medium base contains peptone (pancreatic digest of casein), meat extract (or equivalent), yeast extract, sodium pyruvate, glycine, lithium chloride and bacteriological agar, providing nutrients while suppressing competing microflora.
After sterilization, the base must be supplemented aseptically with egg-yolk emulsion and tellurite (or selective supplement as per protocol) to obtain full selective and differential properties. When inoculated and incubated according to ISO/EN standards, coagulase-positive staphylococci develop characteristic black or grey-black, shiny, convex colonies often surrounded by a clear zone (due to lecithinase activity), facilitating presumptive identification. For full confirmation, coagulase test is recommended.
